Analysis of the production of L929 cell-based cytokine, M-CSF, using 3D spheroid culture system

Abstract

A mouse fibroblast cell line, L929, produces the Macrophage colony-stimulating factor(M-csf) which is a hematopoietic growth factor that plays a critical role in the survival and differentiation of hematopoietic cell lineage.[1] In order to supply sufficient M-csf, three conventional methods are available: co-culturing fibroblast, purchasing recombinant M-csf, and applying M-csf conditioned media. However, conventional methods based on 2-D possess severe drawbacks in space limitation, long term culture, and inconsistent M-csf production. To overcome such inefficiency, we introduce simple spheroid based M-csf harvesting method. Comparing with 2-D data, spheroids numbered at 361 ensure space control, long term culture and M-csf production.
