Detection of post-translational modifications by comparison of intact proteins with tryptic digests in a complex biological sample

Abstract

Post-translational modifications were detected by comparison of intact proteins with tryptic digests in a complex biological sample. Bicinchoninic Acid (BCA) protein assay was utilized to determine protein concentration of samples for intact protein analysis. Final concentration was determined by BCA assay. The intact protein masses were examined to determine possible phosphorylated proteins that corresponded to proteins identified through enzymatic digestion. It was found that only a subset of proteins present in the sample was used to identify the intact protein masses, which reduces the number of mistaken identifications.
