Evaluation of brain-blood barrier permeable peptide using microfluidic reconstituted BBB model

    Abstract

    The Blood-brain barrier (BBB) is a selective barrier blocking the passage of most molecules for the central nervous system (CNS). Because of unique function of blood-brain barrier, the development of therapeutic drugs for brain diseases and evaluating of the CNS drug candidates has been interrupted. To develop a useful in vitro BBB model, we constructed a microfluidic BBB model. We compared the permeability value of hCMEC/D3 monolayer with hMVEC monolayer. Furthermore, we applied a cell-penetrating peptide, Angiopep-2, to our BBB platform. The higher permeability value of Angiopep-2 than same sized dextran molecule was proved in BBB device.
